ATAC-Seq Kit常見問題
1、此試劑是否和Buenrostro et al在nature上發表的方法一樣？
Active motif 基于最初Buenrostro 的文章和2017年Corces et al. 在nature上發表的文章，優化了緩沖液以滿足跟多的實驗需求和高效的實驗過程。
2、試劑盒中包含的重組Tn5酶的來源是？
此試劑盒中的Tn5是由我司克隆，表達，純化，驗證的超活性突變克隆。所提供的這款轉座酶預先已鏈接好用于NGS測序的接頭（adaptor）序列，所以它不許任何預處理過程，可直接用于實驗。
3、每個反應需要使用的小的細胞數量或者組織量是多少？
50,000個細胞或20 mg組織樣本。
4、可以使用凍存的組織和細胞嗎？
可以，但樣本得保證高質量且高活性，細胞應在控制速率冷凍中冷凍保存（例如細胞凍存液梯度凍存），冷凍介質的配方應防止冰晶形成和隨后的細胞損傷，組織應在-80°C下快速冷凍。
5、可將細胞核凍存后用于后續ATAC-seq實驗嗎？
可以的，提取的細胞核可以-80℃凍存用于后續使用。制備的同樣品細胞核可合并在一起增加樣本量。
6、可以使用FACS分選后的細胞嗎？
可能可以。但FACS后細胞可能會被破壞。一個高質量的ATAC-seq 文庫需要高質量高活性的細胞。
7、您是否建議使用40微米篩孔的細胞樣本過濾器？
是的，我們推薦使用Falcon 40微米過濾器（Falcon貨號：352340）。
8、所有樣本都需要DNase處理嗎？
DNase的處理能夠幫助確定懸液中游離DNA的去除，如線粒體DNA的去除，如果細胞樣本中有15%的死亡率，細胞活性不夠且必須使用此酶，它最終可能會移除你想要的細胞數據。只有當細胞是活較高時，并且不會被酶消化時，才建議將其作為一種選擇。
9、若沒有thermomixer，是否還能使用此試劑盒？
建議使用Thermomixer。然而，我們已經對此進行了測試，在37°C溫度下不使用thermomixer而是在帶熱蓋的熱循環器中進行轉座反應，反應能夠成功。
10、 是否能夠室溫融化此試劑盒包含的所有試劑盒組分，如master mixes和酶？
除了轉座酶和Q5 DNA polymerase 以外，其他試劑均可室溫融凍。轉座酶和Q5 DNA polymerase 包裝中含有甘油，不會被凍結，請直接放在冰上使用。其他組分可融凍多次用于以后使用，所以不需要一次性將16次的試劑用完。
11、 每個樣本需要測序多少量的reads？
常規20 M reads的測序量就足夠了。
